Stratford Upon Avon is about 2 hours drive from Central London. So travelling is going to take a good chunk of your day. Your choice is really whether to spend all your time in Stratford or make it a long day and combine it with somewhere else.

Shakespeare's Stratford Upon Avon
The 'must see' is Shakespeare's Birthplace in the middle of town.
On the far side of the town centre is Holy Trinity Church where Shakespeare is buried.
The town itself is also a nice place to loiter with some olde worlde charm and a very nice riverside with extensive parkland and always something going on.

A mile or two from the town centre is Anne Hathaway's Cottage and further out is Shakespeare's mother's house, Mary Arden.

If you're really into Shakespeare you could spend the day here, but your visit can be tailored to your interest.

Combinations
Warwick Castle is only about 30 minutes from Stratford. Unlike Windsor Castle or most other castles open to the public, Warwick has more of an emphasis on entertainment. It is run by the company who run Madame Tussaud's.

The Cotswolds is just south of Stratford and has perhaps the finest countryside accessible in a day trip from London with many examples of the quintessential English Village.
